The Activator (Ac) element is a class II transposable element that is found worldwide and is associated with gene movement. The element has been used for studies of gene tagging in maize as well as in Arabidopsis thaliana. In this study, we cloned a novel maize Ac variant and showed that it could transpose in Arabidopsis. This variant, called Ac-im, had five bp of the left end of the Ac element deleted, as well as a missense mutation in the G-rich region. In addition, Ac-im was capable of activating multiple Ds insertions at the r1 locus in maize. Finally, we discuss the utility of Ac-im for the development of two-component gene-tagging systems in maize using a single Ac-im construct.

We have characterized the orthologous genes of the Pea early browning virus-induced gene 2 (Pea2) in tomato and pepper. A comparison of Pea2 and its orthologs in two tomato cultivars revealed differences in the level and timing of expression and the architecture of the genomic region encoding this protein. Three classes of tomato Pea2 orthologs were identified. One of these had a catalytic triad like those of the tomato MASA-domain proteins, three had a catalytic triad like those of the MADS-domain proteins, and one had a catalytic triad like those of the MADS-domain proteins. These four types of Pea2 orthologs are present in a variety of species of the Solanaceae. The evolution of these proteins and the divergence of their catalytic triads is discussed. PMID:16644920
